Isothiocyanates used with lapatinib can increase the drug’s effectiveness against breast cancer cells to reduce the risk of metastasis, a new study suggests.

Lapatinib is a tyrosine kinase inhibitor that interrupts the HER2/neu and epidermal growth factor receptor (EGFR) pathways. This drug is used in combination therapy to treat HER2-positive breast cancer or advanced or metastatic breast cancer whose tumors overexpress HER2.

A common problem with lapatinib is that long-term exposure to the drug can increase the risk of cancer cells becoming resistant to the drug, leading to a loss of the drug’s effectiveness against breast cancer cells.

The study authors tested lapatinib treatment with one of three types of isothiocyanates, sulforaphane, erucase and sulforaphene, to see how the isothiocyanates affect the drug’s response to lapatinib. Efficacy of Ni-sensitive and lapatinib-resistant breast cancer cells.

In the experiments, both drug-resistant and sensitive breast cancer cells were present in culture but in different proportions. Studies have shown that lapatinib with any isothiocyanate significantly reduces cell viability and inhibits migration of two types of breast cancer cells.

In cultures of only drug-resistant breast cancer cells, combination treatment with erucase or lapatinib most effectively reduced cell viability and motility.

The authors concluded that isothiocyanates, especially carotene, could be used to enhance the effectiveness of lapatinib in breast cancer to reduce the risk of metastasis, even when breast cancer cells are resistant to lapatinib.

Isothiocyanates are found high in Brussels sprouts, garden cress, mustard greens, turnips, savoy greens, and kale. Many other vegetables such as watercress, red cabbage, broccoli, horseradish, cauliflower, and bok choy also contain isothiocyanates.
